RK-Spectro Technical Note 57: Single wavelength kinetic using J715-720 and Bio-Kine in fluorescence mode

Introduction

This technical note demonstrates how to perform single wavelength kinetics using J-715-720, a stopped-flow module and Bio-kine32. The scenario pressuposes that the system has been correctly installed and that a 320 nm cutoff filter is used for the detection.
Reaction used : refolding of Hen egg lysosyme. The enzyme unfolded in a 6M GnDHCl solution is refolded by a 9 fold dilution in water. This test can be performed with all our model of stopped-flow system, for reasons of simplicity, this experiment is described with an SFM-300.
